When diagnosing tuberculosis, many parameters must be taken into account (X-ray, clinical signs, etc. ). However, the direct detection of the pathogen of Mycobacterium tuberculosis from the sputum (sputum produced when coughing) of a patient is still regarded as the key investigation. After staining, the acid-resistant rods can be visualized well under fluorescence microscopy or light microscopy at 1000x magnification.
